7kb plasmid
Abstract
The 7Kb plasmid is a small plasmid found within the species diplococcus aquaticus. This plasmid is found and isolated from grown cultures, from there the use of different techniques are used and then become linear. Then change of a portion of the plasmid will change, then will be reintroduced into the same species.Introduction
Through the process of our experiment, the frit was found to have collected a positively charged molecule within our sample. Having found that the gels previously run were filled with about a tenth of the sample allegedly put into the wells. As this was found using the fluorometer to show that the DNA of 164 ng/ul was found to have only about 11.7ng/ul. From this, it was placed on a gel and there was a band at the 7kb on the ladder. This indicated that even though the concentration is low and the purity is good, there is in fact a band that was shown.
Methods
The previous plasmid extractions were done with a thermoscience kit. There is a homebrew plasmid extraction that is being considered.

Discussion
This week was a week of struggle. While there was some break hopefully there will be a purposeful step forward. We have found that the plasmid extraction from the thermoscience kit seems to have trouble with the frit. So for the following week, we hope to use a homebrew plasmid extraction that will allow us to not use a frit at all. I am going to see if there is any correlation between the use of the frit and the appearance of the positively charged molecule. During this homebrew plasmid extraction, we are prepared to loos about half of any plasmid that we can extract.
