7kb plasmid
Abstract
The 7Kb plasmid is a small plasmid found within the species diplococcus aquaticus. This plasmid is found and isolated from grown cultures, from there the use of different techniques are used and then become linear. Then change of a portion of the plasmid will change, then will be reintroduced into the same species.
Introduction
Th home brew plasmid extraction was discarded as the percipitations would take more of the plasmid then we were willing to give. As the primers for this week were in the process of getting here. Worked on the kanomyosin resistant bacterial insert.
Methods
Use of long AMp PCR, the aneling tempperature was changed to 61*C insteed of the median of 62.6*C. THis proved to leave a streak through our gell as shown in figure 1.
Results
Discussion
During this week there were a lot of question about if this was the direction we should be going in. The talk is that the plasmid needs to be annelied at a diffeerent tempurature. This will be performed next week. After the plasmid is linearized, then we will be able to insert the kanomyosin resistance with xbal hangover.
